Sarcina Lutea (Micrococcus Luteus)
Shape and Colony Characteristics-
is a spherical bacteria that is yellow when grown nutrient agar. Its colonies are tetrad, diplo, or staph.
Ideal Growth Conditions-
S. Lutea grows best at 25 degrees Celsius, which is about 77 degrees Fahrenheit.
Gram Stain Test-
The Gram Stain test on S. Lutea came out positive. That means S. Lutea has a single cell wall layer of carbohydrates and proteins. The bacteria came out as purple in the microscope, which means it absorbed Crystal Violet, the dye soaked up by Gram Positive bacteria.
Methyl Red Test-
The purpose of this test was to see whether S. Lutea produced acid waste or basic waste when it digested glucose broth. After mixing the bacteria in the glucose broth, we incubated it for 48 hours. If the acteria digested the glucose and made acidwaste, it would come out red. If the baccteria made basic waste, the color would be orange or yellow. S. Lutea digested the broth but didn't make acid waste since the broth in the tube was yellow from basic waste.
Purpose and Results of Motility Test-
The purpose of the motility test is to see if the bateria is motile. After stabbing the bateria in a transparent purple agar, we incubated it. The agar was still clear, which means that
Hemolysis Test Explanations-
After placing S. lutea on sheep's blood agar and incubating it, I found that S. Lutea is Gamma. That means that the bacteria didn't digest any of the hemoglobin found in the sheep's blood agar.
After being placed in an air tight jar for twenty four hours, S. Lutea showed no signs of growth. This goes to show that S. lutea is dependent upon oxygen and needs it to grow.
Four antibiotics were tested on S. Lutea. Penicillin showed no signs of effectiveness against S. Lutea. Tetracicline was somewhat effective, with a 24 mm diameter zone of inhibition. Chloriamphenicol had a zone of inhibition of 34 and hand sanitizer had a diameter of 16 for the zone of inhibition. Of the four antibiotics, Cholriamphenicol was the most effective.
S. Lutea is often found in soil, dust, water, and air, as well as beer and some dairy products. S. lutea has also been known to live in inorganic nitrogen and is often found on human skin. It is found in various parts of the body and is one of the culprits responsible for body odor.
Illnesses Associated with Sarcina Lutea-
Though S. Lutea is usually harmless, it has been known to cause skin infections in those with weak immune systems, such as HIV patients.
